Abstract:
The extraction process conditions of Eucommia ulmoides gum using natural deep eutectic solvents(NADES) pretreatment were investigated.Based on single-factor test, orthogonal test and response surface test were employed to optimize the process. The results indicated that when NADES was composed by choline chloride/glucose with molar ratio of 1/2,the pretreatment temperature had the greatest effect on extraction rate of Eucommia ulmoides gum, followed by the material-to-liquid ratio and pretreatment time. The optimal pretreatment temperature obtained from the response surface test was 103 ℃,the material-to-liquid ratio was 1/22 g·mL-1,and the pretreatment time was 2.3 h,and the extraction rate of Eucommia ulmoides gum was 13.73% in verification test.On this basis, reflux extraction process was further optimized, and the best extraction time was 4 h,the extraction cycle was 1,and the solid-to-liquid ratio was 1/25 g·mL-1.Under the combined optimized conditions, the overall extraction rate of Eucommia ulmoides gum reached 14.85%.NADES pretreatment coupled with reflux extraction could significantly enhance the extraction efficiency of Eucommia ulmoides gum, providing reference for its efficient development and utilization.
